c-Jun N-terminal kinases (JNKs) belong to mitogen-activated protein kinase (MAPK) family of proteins and they are known to regulate cell proliferation, invasive/migration, cell survival mediated cancer therapeutic resistance, and various forms of cell deaths (autophagy, apoptosis, necroptosis, ferroptosis, pyroptosis etc.). JNKs are stress activated protein kinases (SAPKs) which can be activated via stress signals, growth factors, inflammatory cytokines, radiation, and other extracellular signals. JNKs-mediated signaling involves a tri-level kinase signaling program that consists of a membrane proximal MAP3K/MAPKKK, a middle MAP2K, and down-most activated JNKs which translocate to the cellular nuclei to regulate multiple transcription factors.
